The rear nut should have a "U" shape to it that fits against the holding bracket, which prevents un-screwing from vibrations. If you have just a flat normal nut, it may be a replacement, you'll need to order a proper piece.

It may be that when you were tightening it the brake line itself was being twisted, so when you pull the brakes the line untwists and loosens. Try to hold the screw part of the line with needle nose pliers or something similar while you tighten the u-bolt.

As far as the brake light. Check the switch at the handle and see if it is putting out 12 volts. Odds are, if it starts with that handle depressed, it's not the switch. Get a multimeter and set it to ohms andground one end to the batterbattery and the other to the inside of the light socket. If the meter goes to 0.00, you have ground. Switch the meter to dc 200 and hit the brake and see if you have 12v on the other lead

You could have stripped the threads out on either the bolt, or the nut. You may have to replace the cable assembly back there.

as far as the brakes go i'd say you are winding up the cable,,you need to hold the metal tip then adjust,,,,the light situation may be caused by any number of things,,,i think you are correct tho with the ground,,check all the grounds to frame connections

I have to hold the threads behind the nut , so the whole thing doesn't turn.

The power wire back there for the brake lights is a green and yellow wire. Easy enough to find. Test it with a meter to see if you are getting power when you squeeze the brake handle. You can tape, or tie the handle down. If you are getting 12v, then you just have to make sure the ground wire is grounded. It should be a solid green wire. There are 3 wires back there. The brown one(yours may be yellow) is +12v for the running tail light, the green and yellow is the +12v for the brake light, and the other one, probably light green(or darker green), is the ground. If you are getting 12v to the green and yellow wire, then you just need to make sure the ground wire runs to the frame somewhere, uninterrupted. You can run a new wire, if you need to. It doesn't need to be anything heavy duty for the brake light. You could also run a wire from the negative terminal on your battery, if you don't see a spot on the frame to attach it to.
