I was just wondering if while I was tuning up a bike and fixing it up and everything that if I didn't tighten the seals completely tight would it mess up the air to gas mixture to much to where it would cause a problem as in wont start or get enough power?
As for example I am always taking off the carb when working on one of my bikes and when I put it back onto the cylinder I dont tighten the connection from the carb to the cylinder, Could that be what would mess up a air to gas ratio?

If you are putting the carb back on and not tightening it, then yes it would not have a proper seal. You would be able to start it but would have running issues.
If it doesn't start you probably forgot to connect something. It can take a few tries to start it after removing carb, because it needs to fill with gas.

It absolutely does affecct A/F ratio, and in a bad way!If more air is getting in than there should be,(and not only from the carb being loose, but improperly connected or leaking vacuum lines or airbox connection),It leans out the mixture thereby burning way hotter in the chamber, and risking damage to the top end.
