Just purchased a new carb and it has a vacuum line attached to it which I think should go to the intake manifold... BUT after removing the hose, that port is blocked off. so my question is; should it be? and if so can I just block off the vacuum port of the intake manifold as I am not using a vacuum powered fuel shut off? This is replacing an EFI so I don't have an OEM carb to compare it to

I'd block the extra port on the manifold and try it. Quality Dept in china isnt great but i dont think its bad enough to totally miss milling an orifice in a casting...

It should go to the fuel line junction after the fuel filter, it is intended to generate vacuum, if you eliminated the vacuum petcock and installed a manual shutoff valve, you can plug it as well as the intake manifold nipple, just leave the fuel line straight from the tank, to the filter, to the carb fuel intake.
