I have a 50cc gy6 with a rear drum cable operated brake. It is possible to put a hydro brake back there and also install a rear rim that accepts disk? I

what you might want to do is find one with disk and analyze what is different.

Indeed, it is possible. Is it worth the effort though?
Remember, most of your stopping power is up front anyway. Sure it will make a difference but, may not be needed in the long run. JMHO
Your money and time is valuable.... Granted it would be a cool project but, (like adding a turbo) The cost and benefit needs to be evaluated. There is a reason why rear drum is so popular.
Good Luck and Ride Safe !!!!!!
